CFTR Modulates Hypothalamic Neuron Excitability to Maintain Female Cycle

Cystic fibrosis transmembrane conductance regulator (CFTR), known as an epithelial Cl− channel, is increasingly noted to be expressed in the nervous system, although whether and how it plays a role neuronal excitability unclear. Given association of CFTR with fertility, we tested here possible involvement regulating hypothalamic neuron excitability. Patch-clamp Ca2+ imaging showed that pharmacological inhibition evoked electrical pulses spikes primary rat neurons, which was dependent on extracellular Cl−. Hypothalamic neurons brain-slice preparations from adult female mice mutation (DF508) exhibited significantly reduced compared wild-type controls. Removal eliminated brain slices, reversible by subsequent addition In mice, indicator (GCaMP6s)-based fiber-photometry activities vivo were enhanced at proestrus/estrus phase diestrus cycle. Such estrus-associated largely diminished DF508 together delayed puberty disturbed cycles. Therefore, these findings suggest critical modulating excitability, may account for cycles fertility associated mutations.
